Glioblastoma multiforme is one of the deadliest cancers with 30 years of very little progression in its treatment and prognosis. In addition to all complexity that is associated with cancer, Glioblastoma is a multiform with multiple morphologies, genetic variations, alteration and appearances. It can either arise from a metastatic cancer in the body or as a primary tumor of the brain. Palladin has been shown to have different roles regarding metastasis and invasion. In some studies, it promoted invasion and in others it functioned as a negative regulator of invasion. Moreover, it can affect tumorigenesis through a possible cross talk with rho GTPases such as Cdc42 and Rac1 that have closely related morphology. In this study, we investigated the roles of palladin and Cdc42 on migration in distinct GBM cell lines. Our results show that palladin played opposite roles in terms of migration in the different cell lines. Moreover, we found out that in A172 calls palladin negatively regulates migration and Cdc42 expression suggesting the possible cross talk between the two proteins and a role of this crosstalk in migration. On the other hand, in SNB-19 cells both palladin and Cdc42 seemed to be positive regulators of migration and no cross talk took place. |
